Perryville began as the county seat of Perry County. In 1821, acreage was deeded to the county. Some of the lots were sold to help fund the building of the first courthouse. There have been three courthouses in Perryville with the current one built in 1904. Perrville was incorporated in 1837, but the incorporation was not kept. Later, in 1856, it was incorporated once again.

Seminary Picnic held every August and the Mayfest each Mother's Day weekend around the city square with entertainment, crafts and fun for all are highlights during the year in Perryville.
